The central redox coenzyme of metabolism and the obligate substrate for sirtuins, PARPs and CD38. A coenzyme, not a peptide.

NAD+ cycles between oxidised and reduced states to carry electrons through glycolysis, the TCA cycle and oxidative phosphorylation. Separately it is consumed as a co-substrate by the sirtuin deacetylases, the PARP DNA-repair enzymes and CD38, which is why intracellular NAD+ availability links mitochondrial function to the DNA-damage response. That dual role, redox carrier and signalling substrate, is the reason it appears across both metabolic and longevity literature.
